RESUMO

<p><b>OBJECTIVE</b>To explore the clinical and genetic characteristics of a case with Pallister-Killian syndrome (PKS).</p><p><b>METHODS</b>Chromosomal karyotype of umbilical cord blood sample derived from a 36-year-old pregnant woman was analyzed by G-banding analysis. After birth, the child was further analyzed with single nucleotide polymorphism microarray (SNP array) and fluorescence in situ hybridization (FISH) using 12pter/12qter probes.</p><p><b>RESULTS</b>G-banding analysis showed that the fetus has a karyotype of 46,XY [77]/47,XY,+mar [23]. After birth, Affymetrix CytoScan 750K array analysis showed a segmental tetrasomy of arr [hg19] 12p13.33p11.1(173 786 - 34 835 641)×4 and a 34.6 Mb repeat at 12p13.33p11.1 with in the neonate. FISH analysis confirmed that 39% of cells harbored the 12p tetrasomy.</p><p><b>CONCLUSION</b>Combined clinical examination, G-banded chromosomal karyotyping, FISH and microarray analysis can delineate the origin and fragments of small supernumerary marker chromosomes and diagnose PKS with precision.</p>

RESUMO

Background:Previous study has found that harmine inhibited the proliferation of gastric cancer cells by down-regulating cyclooxygenase-2(COX-2)expression. However,its molecular mechanism is not fully clear. Aims:To investigate the effect of harmine on proliferation and apoptosis of gastric cancer cells,and explore the role of PTEN/Akt/MDM2 signaling pathway in this process. Methods:Human gastric adenocarcinoma cell line SGC-7901 and MKN-45 were treated with harmine at different concentrations(2,4,8,16,32 μg/mL)for 24,48,and 72 hours. The cell proliferation and apoptosis were detected by MTT assay and Hoechst staining,respectively. The expressions of PTEN,COX-2, phosphorylated Akt(p-Akt)and p-MDM2 were measured by Western blotting. Results:Harmine dose- and time-dependently inhibited proliferation and induced apoptosis of SGC-7901 and MKN-45 cells. Also,harmine dose-dependently increased PTEN expression,and inhibited p-Akt,p-MDM2 and COX-2 expressions in SGC-7901 and MKN-45 cells. Conclusions:Harmine may inhibit proliferation and induce apoptosis of gastric cancer cells via down-regulating COX-2 expression through PTEN/Akt/MDM2 signaling pathway.

RESUMO

Objective To investigate the correlation between serum levels of 25 hydroxyvitamin D and immunoglobulin E(IgE)in adults.Methods There were 1 969 participants over the age of 19 were enrolled in this studied,serum total IgE,dermatophagoides farinae(Df)-specific IgE,and serum 25(OH)D levels were analyzed.Other variables included sex,age,body mass index,smoking history,and economic status were collected.Adjusted regression analysis was used to examine the independent association of 25(OH)D with serum IgE levels.Results The average serum 25(OH)D level of 1 969 patients was(18.0±6.5)μg/L,the prevalence of asthma,allergic rhinitis,and atopic dermatitis were 15.2%(299/1 969),2.8%(63/1 969)and 3.0%(68/1 969)respectively,however,there was no significant difference in serum 25(OH)D levels in patients with allergic rhinitis.All subjects were divided into four groups according to the serum 25(OH)D level: Q1:less than and equal to 13.4 μg/L,Q2:more then and equal to 13.4 μg/L but less then and equal to 17.0 μg/L,Q3: more then and equal to 17.0 μg/L but less then and equal to 21.7 μg/L,and Q4: more than and equal to 21.8 μg/L.The median TIgE level increased with higher quartiles in the Kruskal-Wallis test.The prevalence of Df in Q1,Q2,Q3,Q4 group were 39.8%,33.2%,38.7%and 43.1%,respectively.Linear regression analysis indicated that 25(OH)D levels were positively correlated with lgTIgE(regression coefficient:0.011,95%CI 0.001-0.021),25(OH)D in male patients with dust mite allergy was positively correlated with lgTIgE(regression coefficient:0.026,95%CI 0.010-0.043).There was no significant correlation between 25(OH)D and lgTIgE in the other groups.Conclusion Adult serum 25(OH)D levels is positively correlated with total IgE levels.

Objective To explore the effects of all-trans-retinoic acid (ATRA) on the proliferation of human lung adenocarcinoma cell line A549 and the expression of APLNR (apelin receptor) gene.Methods The inhibition of proliferation of human lung adenocarcinoma cell lines A549 cultured in vitro with or without ATRA was measured by MTT (methyl thiazolyl tetrazolium,MTT) method.The morphological changes in the cells were observed by light microscopy.The cell cycle and apoptosis were analyzed by flow cytometry.The levels of APLNR,cyclin D1 and p16 proteins were detected by western blot.Results After treatment of ATRA,the proliferation of A549 cells was obviously inhibited in dose-and time-independent manner (P < 0.01).The cell morphology was significantly changed.The cycle of A549 cells was blocked at G0/G1 phase and the apoptosis rate was increased.With the increasing concentration of ATRA,the expressions of cyclin D1 and APLNR were down-regulated but the expression of p16 was up-regulated (P < 0.01).Conclusion ATRA could inhibit the proliferation of A549 cells by retardant cell cycle of A549 cells at G0/G1 phase and inducing the apoptosis,and down-regulate the expression of APLNR gene.

Objective To probe into the emergency triage efficiency between emergency rapid triage system (ERTS) and emergency severity classification of 4 level. Methods A total of 46965 emergency patients from January to December in 2015 were selected as the research object and utilized a data of patients from the electronic triage system, and triaged the emergency patients by means of ERTS retrospectively. A comparison was performed in patient visits, outcome and hospital rates between ERTS and emergency severity classification of 4 level. Results In both of these triage systems, almost 80%of patients are assigned to the lowest acuity (levelⅢandⅣ). The visits of patients with gradeⅢand grade IV diseases were 71.5%(33580/46965), 50.5%(23717/46965) in ERTS and 12.1%(5683/46965), 35.1%(16485/46965) in emergency severity classification of 4 level respectively. The outcomes of patient under the ERTS were better than emergency severity classification of 4 level at levelⅠandⅡ(χ2=11.79, 1100.62, P<0.05). At levelⅠandⅡ, ERTS in the patient's disease hospital admission rates were 70.5%(311/441), 72.6%(5274/7264), which higher than emergency severity classification of 4 level, 62.5%(371/594), 45.2%(2785/6161), the difference was statistically significant (χ2=7.32, 1043.13, P<0.05). At level Ⅲ and Ⅳ, ERTS in the patient's disease hospital admission rates were 9.6%(3676/38288), 7.9%(77/972) , which lower than emergency severity classification of 4 level, 19.3%(4578/23719) , 9.9%(1632/16491), the difference was statistically significant (χ2=1153.10, 4.05, P<0.05). Conclusions ERTS significantly improves the sensitivity and accuracy of emergency triage compared to emergency severity classification of 4 level. ERTS has obvious significance in the emergency patient distribution, and also has a certain advantage in the patient outcomes and hospitalization rates. It can be used as a auxiliary tool.

[ ABSTRACT] AIM:To study the expression of miR-203 in tongue carcinoma tissues and the effect of miR-203 over-expression on the viability and invasion ability of Tca 8113 cells.METHODS:Twenty-eight pairs of tongue carcinoma tissues and adjacent nontumor tissues were collected , and the clinicopathological characters were analyzed .miR-203 was detected in the tongue tissues of 28 patients with tongue carcinoma by real-time PCR.miR-203 mimics and scramble were transfected into Tca8113 cells by Lipofectamine 2000.The expression of miR-203 was detected in Tca8113, Tca8113-miR-203 mimics and Tca8113-scramble cells by RT-qPCR.The cell viability was measured by CCK-8 assay.The cell invasion ability was determined by Transwell chamber invasion experiment .RESULTS:miR-203 expression was significantly down-regulated in the tongue carcinoma tissues compared with those in the adjacent nontumor tissues .The expression of miR-203 was associated with TNM stage and lymph node metastasis .Up-regulation of miR-203 inhibited the viability and invasion a-bility of Tca8113 cells.CONCLUSION:miR-203 suppresses the growth and invasion of tongue carcinoma cells .miR-203 may be a potential therapeutic target for treating human tongue cancer .

<p><b>OBJECTIVE</b>To examine cerebral pathologies in cerebral amyloid angiopathy in a rat model of Alzheimer's disease.</p><p><b>METHODS</b>Rat models of Alzheimer's disease was established by stereotactic Aβ1-42 fiber injection in the bilateral hippocampus. The cognitive function of the rats was evaluated with water maze test. HE staining, Congo red staining and double-labeling indirect immunofluorescence were used to examine the dynamic distribution of Aβ fiber deposit in the brain.</p><p><b>RESULTS</b>The model rats showed significant differences from the control rats in the escape latency and the times of crossing platform in waster maze test. HE staining revealed a decreased number and degeneration of the granular cells with increased glial cells in the model rats. Congo Red staining showed that the Aβ fiber was deposited gradually in the small vessels in the brain parenchyma to cause thickening, stenosis or occlusion of the small vessels. Immunofluorescence staining detected Aβ fiber migration from the parenchyma to the walls of the small arteries in the rat models.</p><p><b>CONCLUSION</b>Cerebral amyloid angiopathy is a major pathological feature in Alzheimer's disease.</p>

RESUMO

Long non-coding RNAs (lncRNAs) are non-coding RNAs that have transcript lengths exceeding 200 bp and do not have the capacity for protein coding because of having no open reading frame. In the human genome, lncRNAs play important regulato-ry roles in the process of epigenetic, transcription and post-transcription, so they have become the focus of research followed by mi-croRNAs. The abnormal expression of lncRNAs in cancer usually represents different functions, such as the function of oncogenes or tumor suppressor genes, which promote or inhibit tumor growth. Lung cancer is a common malignant tumor with a five-year survival rate of 17%. Previous literature shows that MALAT1, H19, lincRNA p21, UCA1, and BC200 are closely related to the development of lung cancer. They could promote cancer growth, invasion and metastasis, apoptosis and induce drug resistance, and so on. This review aims to provide assistance in the diagnosis, treatment, and prognosis of lung cancer by clarifying the functions and mechanisms of ln-cRNAs.

<p><b>OBJECTIVE</b>To study the effect of kallikrein-related peptidase 10 (KLK10) on the proliferation and invasiveness of human tongue cancer cell line Tca8113.</p><p><b>METHODS</b>The eukaryotic expression vector harboring KLK10 gene (pIRES2-EGFP-KLK10) was transfected in Tca8113 cells and the stable cell lines were selected by G418 screening. The mRNA and protein expression of KLK10 in transfected Tca8113 cells were assayed by RT-PCR and Western blotting, respectively, and the proliferation and invasiveness of the cells were evaluated by MTS cell growth assay and Transwell chamber invasion experiments.</p><p><b>RESULTS</b>A stable Tca8113 cell line with high KLK10 expression was obtained, which showed significantly increased mRNA and protein expression levels of KLK10 and obviously attenuated proliferation and invasiveness compared with control and empty vector-transfected cells (P<0.05).</p><p><b>CONCLUSION</b>Enhancing KLK10 gene expression can decrease the proliferation and invasiveness of human tongue cancer cells in vitro.</p>

RESUMO

Objective To study the relationship between cleanliness of children′s hands and diminution of Ascaris lumbricoides infection. Methods Before the study all persons positive for ascaris eggs in the preliminary survey were treated with albendazole. Hand washing habit before meal and after defecation was kept in children of experimental group, but not in the control group. Kato thick smear stool examination was done once every two months for one year to compare the new infection rates in children without ascaris infection in the two groups, and the reinfection rates in the cured negative cases were also compared between them in half a month after chemotherapy. Results All the new infection rates as well as reinfection rates of each reexamination in the experimental group were significantly lower than that of the control group ( P
